油菜无菌苗培养前的种子消毒技术

摘  要:
油菜种子浸泡与否对消毒效果的影响研究表明,在种子消毒之前,经无菌水浸泡30h(预处理)的比不浸泡的消毒效果好,无菌水浸泡后再经0. 1%升汞消毒的种子带菌率为40. 7%,而未经无菌水浸泡的带菌率为53. 6%。采用不同杀菌剂恒温下浸种的方法,发现50%地菌杀净可湿性粉剂对油菜种子的内生菌有很强的杀菌作用,处理后的油菜品种M004和甘油5号平均带菌率分别为0%和1. 1%,极显著地低于无菌水对照和0. 1%升汞处理。地菌杀净浸种的温、时效应对种子杀菌消毒效果的影响研究表明,提高浸种温度、延长浸种时间,可增强杀菌消毒效果。用50%地菌杀净浸种的最佳杀菌、消毒处理是地菌杀净药液浓度为1 000mg/L,处理温度为37℃。

摘  要:
The effect of seed soaking on sterilization was studied by soaking rapeseed seeds in aseptic water before seed sterilization. The result revealed that sterilization of seeds soaked in aseptic water for 30 hours was better than that without soaking. The germ-bearing rate of seeds soaked in aseptic water before sterilization with (0.1%) mercuric chloride was 40.7%, while the germ-bearing rate of seeds without soaking was 56.3%. By means of seed soaking with different fungicides under constant temperature, the sterilization effects of fungicides on rapeseed seeds were studied. Results showed that 50% Dijunshajing WP had a great sterilization effect. The germ-bearing rate of seeds of M004 and Ganyou-5 treated with Dijunshajing was 0% and 1.1%, respectively, which was much lower than that of CK and seeds treated with 0.1% mercuric chloride. The temperature and time used for seed soaking with Dijunshajing influenced sterilization. Higher temperature and longer time could enhance the effect of sterilization.It was proved to be the best when seeds soaked in the suspension for 7h, at 37℃.
