黄河鲤figla基因表达分析

生殖系a因子(figla)作为雌性生殖细胞特异性转录因子已被证明对哺乳动物卵泡的发育和透明带的形成具有调节作用,figla基因异常会引起卵巢早衰。然而,figla在硬骨鱼中的潜在角色仍然难以确定。为了确定figla在黄河鲤(Cyprinus carpio)卵巢发育中的潜在功能,本研究中分离黄河鲤figla基因全长ORF序列,通过RT-PCR分析其在不同组织及不同发育时期胚胎中的表达情况。结果显示,黄河鲤figla基因ORF全长672 bp,编码223个氨基酸。氨基酸序列比对分析结果显示黄河鲤与其它物种的figla同源性较低。系统进化分析显示,黄河鲤Figla与金线鲃等鲤科鱼类亲缘关系最近。RT-PCR分析表明,Figla基因仅在黄河鲤卵巢中表达,在其它组织(包括雄鱼)中均不表达。同时,RT-PCR结果表明figla在早期胚胎发育过程中在未受精卵、受精卵以及卵裂期表达。综上所述,在黄河鲤中figla基因作为一个母源因子可能对卵巢发育有重要作用。

摘 要:
Germline factor a(figla), a specific transcription factor for female germ cells, has been proved to have a regulatory effect on the development of mammalian follicles and the formation of zona pellucida. Abnormal figla gene can cause premature ovarian failure. However, the potential role of figla in teleosts remains elusive. In order to determine the potential role of figla in the ovary development of yellow river carp(Cyprinus carpio), the full length ORF of figla gene was isolated and the expression of it in different tissues and embryonic development stages was analyzed by RT-PCR in Cyprinus carpio. The results showed that the ORF of figla was 672 bp in length, encoding 223 amino acids. Amino acid sequence comparison analysis showed that the figla homology of Cyprinus carpio and other species was low. Alignment of the Cyprinus carpio Figla with those from other organisms revealed a low identity. Phylogenetic analysis showed that the figla of Cyprinus carpio clustered with those of fish.RT-PCR analysis revealed that the figla gene was highly expressed in the ovary of Cyprinus carpio, but was not detected in other tissues(including males'). In the different periods during the early stages of development figla was only detected in the unfertilized eggs, fertilized egg and cleavage stage. Taken together, as a maternal factor,figla may play a regulatory role in ovarian development in Cyprinus carpio.
