葛根芩连汤及葛根素对脂多糖诱导小鼠小肠粘膜微血管内皮细胞分泌一氧化氮和内皮素的影响

摘  要:
为探索葛根芩连汤及葛根素对脂多糖(LPS)诱导小鼠小肠粘膜微血管内皮细胞(MIMVECs)分泌一氧化氮(NO)和内皮素(ET-1)的影响,本实验将葛根芩连汤、葛根素分别与LPS加入细胞板中,降低对原代MIMVECs的病理损伤,在不同时间,采用硝酸还原酶法和ELISA检测MIMVECs分泌的NO和ET-1。结果显示:在3 h~12 h,葛根芩连汤实验组与LPS对照组分泌的NO和ET-1的含量差异显著(p<0.05);葛根素实验组与LPS对照组分泌的NO和ET-1的含量差异显著(p<0.05)。表明葛根芩连汤和葛根素均能够对LPS损伤MIMVECs起到拮抗的作用,为进一步研究葛根芩连汤及其有效成分对LPS损伤MIMVECs的机理奠定基础。

摘  要:
In order to explore the effect of Gegenqinliantang and puerarin on lipopolysaccharide(LPS) leading mice intestines microvascular endothelial cells(MIMVECs) to secret NO and endothelin(ET-1), the Gegenqinliantang and puerarin accompanying with LPS were added into cell monolayers, respectively, to reduce the pathologic impact to MIMVECs. The secreting NO and ET-1 from the treated MIMVECs were detected by nitrate reduction method and ELISA. The results showed that the concentration of NO between the experimental group of Gegenqinliantang and control group of LPS were significant different(p<0.05) at 3hrs and 12 hrs, the concentration of ET-1 between experimental group and control group had significant difference(p<0.05) during3 hrs and 12 hrs, the concentration of ET-1 between the experimental group of puerarin and the control group of LPS were also significant different. The results showed that both Gegenqinliantang and puerarin were able to reduce the damage of MIMVECs induced by LPS. These data provided a base for further exploring the mechanism of Gegenqinliantang and its active ingredient hurt MIMVECs.
