摘 要:
以昆明小鼠为实验动物,检测柞蚕雄蛾粉的急性毒性和致突变性,为柞蚕雄蛾药品和食品的毒理学与安全性评价提供试验依据。将柞蚕雄蛾粉按照18 g/kg的总剂量在24 h内分3次饲喂小鼠,然后连续14 d均未观察到小鼠有中毒症状和死亡情况,检测柞蚕雄蛾粉对小鼠经口急性毒性的LD50>15 g/kg,属无毒级物质。雌性和雄性小鼠分别按2.5 g/(kg·d)、5.0g/(kg·d)、10.0 g/(kg·d)3种剂量饲喂柞蚕雄蛾粉,小鼠的骨髓细胞微核率与阴性对照组比较均无显著性差异(P>0.05),与环磷酰胺处理的阳性对照组比较差异显著(P<0.05);3个剂量组小鼠的嗜多染红细胞占红细胞的百分率(PCE/RBC)与阴性对照组比较均无显著性差异(P>0.05),但与阳性对照组比较均有显著性差异(P<0.05)。致突变性(Ames)检测试验中,以16、78、313、1 250、5 000μg/皿5种剂量的柞蚕雄蛾粉处理后,各处理组的回变菌落数与空白对照组、溶剂对照组及组氨酸对照组比较均无显著性差异(P>0.05),与阳性对照组比较均有显著性差异(P<0.05),且5个剂量处理组间无剂量效应关系(无显著性差异)。2.5 g/(kg·d)、5.0 g/(kg·d)、10.0 g/(kg·d)3种剂量柞蚕雄蛾粉添食试验组雄性小鼠的精子畸形率与阴性对照组比较均无显著性差异(P>0.05),与环磷酰胺处理的阳性对照组比较均有显著性差异(P<0.05)。以上试验结果初步说明柞蚕雄蛾粉对昆明小鼠无急性毒性、无致突变性和无致畸性。

摘 要:
In this paper,using Kunming mice as animal model,acute toxicity and mutagenicity of tussah male moth powder were investigated to provide experimental basis for toxicology study and safety evaluation of medicine and food developed from tussah male moth. After feeding Kunming mice with tussah male moth powder at total dosage of 18 g / kg for three times at 24 h,poisoning symptoms and death were not observed during investigation period(14 d). The results of acute toxicity test showed that LD50 of tussah male moth powder to Kunming mice was over 15 g / kg,belonging to substance of non-toxic grade. After female and male mice were fed with tussah male moth powder at the dose of 2. 5g /( kg·d),5. 0 g /( kg·d) and 10. 0 g /( kg·d),the micronucleus rate of bone marrow cells of tested mice was not significantly different with that of the negative control group(P >0. 05),but had significant difference with that of the positive control group treated with cyclophosphamide( P< 0. 05). The percentage of polychromatic erythrocytes in red blood cells( PCE/ RBC) of tested mice in three dosage groups had no significant difference with the negative control group( P > 0. 05),but had significant difference with positive control group( P < 0. 05). Mutagenicity test was conducted by treatment with tussah male moth powder of 16,78,313,1 250,and 5 000 μg per plate respectively. The number of revertant colonie of all dosage groups had no significant difference with blank control group,solvent control group and the positive control group treatment with histidine( P > 0. 05),but had significant difference with positive control group( P < 0. 05). Besides,there was no dose response relationship between five dose goups( no signifcant difference). After fed with tussah male moth powder at the dose of 2. 5 g /( kg·d),5. 0 g /( kg·d) and 10. 0 g /( kg·d),the sperm malformation rate had no significant difference with the negative control group( P > 0. 05),but had significant difference with positive control group treatment with cyclophosphamide( P < 0. 05). The above results indicated that tussah male moth powder had no acute toxicity,mutagenicity and teratogenicity on Kunming mice.
