六月雪水提取物对乙醇损伤的人胃黏膜上皮细胞的保护作用

摘  要:
前期研究发现六月雪[Serissa serissoides(DC.)Druce]对乙醇诱导的大鼠胃黏膜损伤有良好的保护作用,因此进一步在细胞水平上探讨六月雪水提取物对乙醇损伤的人胃黏膜上皮细胞的保护作用。采用体外细胞培养人胃黏膜上皮细胞系GES-1细胞,将GES-1细胞用0~10%的乙醇孵育4~6 h,建立胃黏膜损伤模型。将参试GES-1细胞分为正常组、模型组、荆花胃康胶丸组、六月雪水提取物剂量组,正常组不做任何处理培养24 h,其他组加入一定浓度的乙醇处理一段时间后,加入含荆花胃康胶丸、六月雪水提取物各浓度剂量的培养基,正常组、模型组不加入药物,继续培养24 h,利用MTT检测各组GES-1细胞的活性。将MTT筛选后效果最佳的六月雪3个浓度以及荆花胃康胶丸组1个浓度分别定为高、中、低剂量组和阳性药物组,运用吖啶橙荧光染色法检测各组细胞凋亡情况,采用Western-Blot法检测各试验组碱性成纤维细胞生长因子(b FGF)蛋白的表达情况。结果表明,5%的乙醇孵育GES-1细胞4 h是造成胃黏膜损伤的最佳细胞模型,对GES-1细胞的抑制率达到40%。六月雪水提取物组中,60、70、80μg/m L 3个浓度效果最佳,与该组其他浓度比较差异显著(P<0.05);荆花胃康胶丸组中,在浓度为160μg/m L时细胞增殖最明显;均高于其他浓度的荆花胃康胶丸组;吖啶橙染色后显微镜下观察正常GES-1细胞形态均一、生长良好,而模型组大量细胞明显出现衰老或凋亡,荆花胃康胶丸组、六月雪水提取物组GES-1细胞生长情况明显好于模型组;模型组b FGF的蛋白表达水平明显高于正常组,差异显著(P<0.05);与模型组比较,六月雪水提取物组的b FGF蛋白表达水平明显增加,差异极显著(P<0.01);六月雪水提取物组与荆花胃康胶丸组比较,b FGF的表达水平差异也达极显著水平(P<0.01)。说明六月雪水提取物可以保护乙醇损伤的人胃黏膜上皮系GES-1细胞,可以增加b FGF的蛋白表达水平,提高溃疡愈合速度。

摘  要:
Previous research found that junesnow [Serissa serissoides(DC.) Druce] has protect effect on gastric mucosal damage of rat caused by alcohol, further research was conducted on cellular level to explore the protect effect of junesnow on human gastric mucosal epithelial cells injured by alcohol. Epithelial cells GES-1 was cultured in vitro, then incubated under alcohol stress(0~10%) for 4~6 h to set gastric mucosal damage model. The experimental cell were divided into normal group,model group, Jinghuawenkang group and Junesnow group. Normal group was cultured for 24 h without any treatment; while the rest groups were treated by alcohol for certain time and then cultured with medium adding different dose of Jinghuawenkan or junesnow for 24 h. MTT experiment was used to exam the cell activity of each group. The results were taken to confirm high, meddle, low dosage of junesnow and positive drug group of Jinghuawenkang. Acridine orange staining method was used to measure apoptosis of each group; and Western-Blot to measure expression of b FGF. Results showed that incubating GES-1 in 5% alcohol for 4 h could establish the best gastric mucosal damage model as the inhibitory rate of cell could reach to 40%. In Junesnow group, 60, 70, 80 μg / m L treatments had the best effect(P<0.05). In Jinghuawenkang group, the cell reproduced the most obviously in 160 μg / m L treatment, higher than other Jinghuawenkang treatments. After acridine orange staining, the normal GES-1 cells showed uniform morphology and well growth status; while the model group cells were obviously senescent or apoptosis. The cells of Junesnow and Jinghuawenkang groups grew better than model group.The expression level of b FGF in model group is significantly higher than in normal group(P <0.05). he expression level of b FGF in Junesnow group is very significantly higher than in model group(P<0.01) and Jinghuawenkang group(P<0.01). The junesnow can protect human gastric mucosal epithelial cells injured by alcohol, can increase the expression of b FGF to improve the ulcer healing rate.
