微生物在铬污染土壤中的分布及铬累积菌株的初步筛选

摘  要:
通过现场采集土样及室内培养测定,对铬矿渣堆放区污染土壤的微生物分布进行了分析。结果表明,微生物的数量分布随污染程度的降低而上升,距离中心污染区越远,微生物CFU的数量就越多;地下样品的CFU数量明显低于表土CFU数量,显示铬污染向地下迁移的趋势。从污染土壤中分离到的抗铬微生物,主要为细菌和霉菌。细菌多为革兰氏阳性菌,且以芽孢菌为主。其中有4株细菌抗总铬能力可达1000mg·L-1,并显示出一定的降铬趋势,在100mg·L-1实验条件下,采用二苯碳酰二肼显色法检测结果显示,试验组低于无菌空白对照50%以上。

摘  要:
Microbial distribution in area of chromic slagheap was revealed with samples from surface and 20 cm depth soil, which were sampled separately at 3, 5 and 50 m around the central chromic slagheap. The results showed that microbial population existed in the Cr polluted soil was closely related to the distance from the slagheap, and the number of CFU in sub-layer soil was much fewer than that in the surface soil, indicating that chromic pollution tended to migrate downwards to sub-layer, and the farther the distance from slagheap, the more the CFU, whereas the deeper beneath surface, the fewer the CFU. The CFU ranged from 104 to 105·g-1, much lower than that in the non-polluted soil (usually above 109). In addition, the isolates that were tolerant to chromium in polluted soil were mainly moulds and Gram-positive bacteria. Four of the bacterial strains were identified to be Cr resistant detected by DPCZ reaction assay, showed tolerance to Cr at 1 000 mg·L-1 level. These strains exhibited an erasing tendency with above 50% of the Cr amount (given in form of K2Cr2O7) than that of the CK at 100 mg·L-1 level in laboratory scale.
