摘  要:
采用双向电泳技术(Two-dimensional gel electrophoresis,2-DE)将冻融前后姜曲海猪的精子蛋白进行分离,采用MALDI Tof/Tof质谱鉴定技术对相关差异蛋白进行分析,并采用blast2 go软件对精子差异蛋白质进行GO(gene ontology)功能注释。结果表明:精子冻融前后的42个差异蛋白点发挥7个生物学功能,分别为装配、催化、结构分子活性、调节分子功能、抗氧化、转录和转运等。综合分析NCBInr蛋白数据库和GO功能注释,获得有意义差异蛋白7个,均下调表达(P<0.05);其中,能量代谢相关蛋白3个、结构相关蛋白2个、受精相关蛋白1个和酶相关蛋白1个。表明冻融影响姜曲海猪精子能量代谢、细胞骨架、受精和抗氧化能力。

摘  要:
In this paper,differential proteomics technique was used to explore cryodamage mechanism of Jiangquhai boar sperm.The expressed proteins of sperm before and after freezing were separated using two-dimensional gel electrophoresis(2-DE) technology.The related differential proteins were identified by MALDI Tof/Tof mass spectrometry(MS).Then MS data was analyzed by Mascot.At last,biological information was performed by blast2 go software.Forty-two differential expressed proteins were found.They performed 7 biological functions,involving binding,catalytic,structural molecule activity,regulation of molecular function,oxidation resistance,transcription and transport.According to the result of NCBInr protein database and GO analysis,7 proteins were identified as meaningful differentilal expressed proteins.They were all down-regulated(P <0.05).These proteins could be divided into 4 functional categories,including energy metabolism(3 proteins),structure maintaining(2 proteins),fertilization(1 protein) and antioxidant(1 protein).In brief,freezing and thawing have effects on energy metabolism,eytoskeleton,fertilization and antioxidant capacity of Jiangquhai boar sperm.
