德宏水牛及杂交后代FSH-β基因多态性与繁殖性能的关系

摘  要:
[目的]本研究分析德宏水牛及杂交后代FSH-β基因多态性,为了解水牛繁殖性能的分子基础及建立检测分子标记奠定基础。[方法]采用PCR及直接测序法检测了德宏水牛及杂交后代FSH-β基因的多态性,并分析了该基因变异位点不同等位基因与繁殖性能的关系。[结果]德宏水牛及杂交后代80个个体的FSH-β基因序列共检测到27个SNP位点,未发现插入/缺失变异。其中2个位于第一内含子,3个位于第二外显子,13个位于第二内含子,6个位于第三外显子,3个位于3'非转录区,9个外显子变异中5个为引起氨基酸改变的功能性变异。经χ2检验,水牛FSH-β基因全部27个变异位点都处于群体遗传平衡状态。而在所有27个变异位点中,有23个位点的等位基因频率在繁殖良好和繁殖异常两个群体中存在差异。[结论]FSH-β基因在德水牛及杂交后代具有很高的基因变异性,可能是影响水牛繁殖性能的一个主要基因。

摘  要:
【Objective】In order to understand molecular basis of reproduction and establish genetic markers in domestic water buffalo,FSH-βpolymorphisms were investigated in DeHong buffalo and hybrids.【Method】Polymorphism of FSH-βgene in DeHong buffalo and their hybrids were investigated by direct sequencing and PCR.And we analyzed the relation of different allele of variable sites and reproductive performance in FSH-βgene.【Result】Total of 27 SNPs were found in sequence of FSH-βgene from 80 DeHong buffalo and their hybrids,no insert and deletion variation were found.In all SNPs,two were in intron 1,three were in exon 2,thirteen were in intron 2,six were in exon 3 and three were in 3'-UTR region.The five out of nine exon SNPs were functionality variation with resulting amino acids change.All 27 SNPs of FSH-βgene in buffalo population were of Hardy-Weinberg equilibrium by chi-square test.In all 27 SNPs,allele frequency of 23 SNPs existed differences between eugonic population and reproduce abnormal population.【Conclusion】FSH-βgene was of high polymorphism in Dehong and hybrid buffalo population,which may be a major gene influencing reproductive traits of buffalo.
