反义磷脂酶Dγ基因转化悬铃木的研究

摘  要:
我们已经建立了利用悬铃木叶片培养诱导植株的高频再生体系。本研究是在这一体系的基础上,以继代生长40天左右的组培苗顶部充分展开的叶片为受体,用含有反义磷脂酶Dγ(PLDγ)基因的农杆菌浸染叶片,通过在含有卡那霉素的培养基上进行抗性筛选,获得了抗性植株。对抗性植株进行了PCR和PCR-Southern杂交鉴定,证明反义PLDY基因已整合进悬铃木核基因组中。

摘  要:
We have previously established The high frequency regeneration system by using the young leaves of Platanus acerifoliaWill as the culturing materia.Basing on this regeneration system,the ndedleaves-8ultured about 40 day were used as the materials for gene transformation.The Agrobacteria tumerference containing pKYLX7 plasmid harboring with antisense-PLDγ gene were used to infect the young leaves.Many kanamycin resistant plantlets were obtained by culturing the infected leaves on the selection medium added with kanamycin.PCR and PCR-Southern tests showed that the antisense-PLD γ gene has been integrated into Platanus acerifoliaWilld genome.
