类金属砷螯合物人工抗原的合成与鉴定

摘  要:
为了合成砷元素人工免疫抗原及人工鉴定抗原,制备砷的多克隆抗体,建立ELISA快速检测方法,以异硫氰酸苄基乙二胺四乙酸(ITCBE)为螯合剂,将砷与载体蛋白BSA和OVA偶联,制备人工免疫抗原As~(3+)-ITCBE-BSA和人工鉴定抗原As~(3+)-ITCBE-OVA;经核酸蛋白仪浓度的检测、紫外分光光度计扫描以及SDS-PAGE定性鉴定,用原子荧光光谱法检测抗原中As~(3+)的含量。结果显示:合成的As~(3+)-ITCBE-BSA和As~(3+)-ITCBE-OVA蛋白浓度分别为4. 692和5. 726 mg/mL;紫外扫描最大吸收峰发生偏移,电泳条带对于各自载体蛋白稍微滞后,均显示抗原合成成功; As~(3+)的含量分别为0. 254、0. 190 mg/mL,偶合率达到70. 67%和52. 81%。提示:抗原合成成功,可用于重金属砷多克隆抗体的制备。

摘  要:
This study was to synthesize artificial immune antigen and artificial identification antigen of the arsenic element,so as to prepare a polyclonal antibody of metal arsenic and to develop a rapid detection method with ELISA. The arsenic element was coupled to the vector proteins,bovine serum albumin( BSA) and ovalbumin( OVA),via a bifunctional chelating agent isotrhiocyanobenzyl-EDTA( ITCBE) respectively,in order to prepare an artificial immune antigen As~(3+)-ITCBE-BSA and an artificial identification antigen As~(3+)-ITCBE-OVA. The concn was measured by nucleic acid protein analyzer,ultraviolet scan of spectrophotometer and SDS electroporesis qualitative indentification.The content of arsenic ions in antigen was determined by the AFS method. The results showed that that the protein concns of As~(3+)-ITCBEBSA and As~(3+)-ITCBE-OVA were 4. 692 mg/mL and 5. 726 mg/mL,respectively,and that the maximum scanned UV absorption peaks shifted. Compared with vector proteins,the synthesized antigens exhibited lagged protein bands in an SDS-PAGE pattern. In general,the synthesis of the antigens was successful,with the concentrations of As~(3+)being 0. 254 mg/mL and 0. 190 mg/mL,respectively,and the coupling rates being as high as 70. 67% and 52. 81%,respectively. The synthesized antigens might be used for preparing the polyclonal antibody of metal arsenic.
