摘 要:
对栝蒌脱毒苗组培快繁技术体系进行了研究。剥取0.2 mm大小经人工接种CMV的栝蒌植株茎尖培养,经RT-PCR检测,能近100%的脱除CMV,获得了脱毒栝蒌苗。以脱毒栝蒌苗带腋芽的茎段为材料,于MS+6-BA 2.0 mg/L培养基中培养,可使栝蒌以侧芽丛生方式实现继代和增殖;3叶以上的栝蒌小苗,采用1/2MS+NAA 0.2 mg/L配方生根培养基,可使栝蒌产生健壮根系,20~30℃移栽于珍珠岩基质苗床,成活率高于90%。剪取苗床中3~4叶侧枝或茎尖扦插于珍珠岩苗床,生根率>90%,可大幅度降低脱毒苗扩繁费用。1 a以上大田栝蒌侧枝或顶芽扦插成活率极低,不能进行扦插繁殖。大规模生产试验表明,脱毒繁殖的栝蒌种苗在生长势、单株结果数等方面呈现明显优势。

摘 要:
The tissue culture and rapid propagation system for virus-free seedlings of Trichosanthes kirilowii were studied.0.2 mm of shoot tip culture of Trichosanthes kirilowii artificially infected CMV was detected by RT-PCR,it found out the virus-free rate of CMV approached to 100%,and the virus-free seedlings of Trichosanthes kirilowii was obtained.The virus-free seedling of Trichosanthes kirilowii with axillary bud was taken as material and cultured in the culture medium of MS+ 6-BA 2.0 mg/L,the results showed that this medium can make Trichosanthes kirilowii proliferated as tufty axillary bud;the culture medium of 1/2 MS+NAA 0.2 mg/L can make small seedlings of Trichosanthes kirilowii with over three leaves rooting strong,and when transplanted the rooted seedlings of Trichosanthes kirilowii to perlite seedling-bed at 20~30℃,the survival rate reached 90%.The side-branch of Trichosanthes kirilowii with three or four leaves or shoot tip of Trichosanthes kirilowii plant in the perlite seedling-bed,the rooting rate will higher than 90%,and the propagation cost of virus-free seedlings will be reduced greatly.Over one-year-old side branch or shoot tip of Trichosanthes kirilowii plant in the field have extremely low survival rate,so they can not use to propagation.The results of mass production experiment indicated that virus-free propagation seedlings of Trichosanthes kirilowii showed superiorities in the growth vigor and the fruit setting rate per plant.
