山蜡梅愈伤组织诱导与植株再生

摘  要:
以山蜡梅下胚轴为材料,研究了去子叶胚萌发、下胚轴愈伤诱导、不定芽分化以及生根的方法.结果表明:去子叶胚最佳萌发培养基为WPM+1.0 mg·L-16-BA,萌发率达76.67%;下胚轴最佳愈伤诱导培养基为WPM+2.0mg·L-12,4-D+0.1 mg·L-1IBA,愈伤诱导率为100%;WPM+0.01 mg·L-1KT+0.01 mg·L-1生物素D+0.05 g·L-1水解酪蛋白较适合诱导不定芽分化,分化率为45.83%,平均不定芽数3.36个;WPM+1.0 mg·L-1NAA诱导生根最佳,最高生根率可达45.0%.

摘  要:
The hypocotyls of Chimonanthus nitens were used to study the germination of decotyledon, callus induction of hypocotyls, adventitious bud differentiation and rooting. The results indicated that the optimal germination medium for decotyledon was WPM +1.0 mg·L~(-1)6-BA, with a germination rate of 76.67%. The optimal callus induction medium was WPM + 2.0 mg·L~(-1)2, 4-D + 0.1 mg·L~(-1) IBA, and the callus induction rate was 100%. WPM + 0.01 mg·L~(-1) KT+ 0.01 mg·L~(-1) D-Biotin + 0.05 g·L~(-1) Hydrolyzed casein is more suitable to induce adventitious bud differentiation, with a differentiation rate of 45.83% and an average number of adventitious buds 3.36. WPM + 1.0 mg·L~(-1) NAA was the best to induce rooting, and the highest rooting rate was up to 45.0%.
