量子点制备和表征及标记黄曲霉毒素B_1性能分析

摘  要:
用碲粉、水合氯化镉和巯基丙酸在水相中合成了发光量子点碲化镉纳米晶标记物,通过TEM和荧光分光光度计对其进行表征;Cd Te量子点与羊抗兔Ig G相连接,对其进行电泳表征,此复合物即为荧光显示探针。将人工抗原AFB1-BSA包被于96微孔板上,黄曲霉毒素B1(AFB1)为模式分析物,与黄曲霉毒素单克隆抗体发生竞争反应。之后用荧光显示探针与AFB1单克隆抗体进行特异性免疫,以量子点探针的荧光强度定量测定AFB1。在适宜试验条件下,AFB1浓度在1~1 000 ng/m L范围内,荧光强度与AFB1含量成线性关系,检出限为1ng/m L。

摘  要:
We used Te,hydrated Cd Cl2 and thiohydracrylic acid to prepare Cd Te QDs as a marker,and characterized them by TEM and fluorospectrophotometer,then we coated avidin onto QDs and conducted electrophoresis and TEM characterization, the compound should be fluorescent display probe. Then,we coated the artificial antigen AFB1-BSA onto 96-microwell plate. As a model analyte,AFB1 had competitive responses with aflatoxin Monoclonal antibody. Then,we used fluorescent display probe to specific immune with AFB1 Monoclonal antibody. We detected the quantity of standardized AFB1 by fluotescence intensity. Under the optimistic conditions,the fluorescence intensity was linear over the range 1 ~ 1 000 ng / m L of standardized AFB1 with a detection limit of 1 ng / m L.
