酸性染料比色法测定苦马豆中总生物碱含量

摘  要:
采用酸性染料比色法测定苦马豆[Swainsonia salsula Taubert(Sphaerophysa salsula(Pall.)DC.)]中总生物碱含量。以苦参碱为标准品,溴麝香草酚蓝为染色液,在p H 7.6缓冲溶液中与生物碱反应,反应产物用氯仿萃取,萃取液于414 nm处测定其吸光度。应用建立的酸性染料比色法对宁夏不同地区采集的苦马豆样品中总生物碱含量进行分析。结果表明,苦参碱在0.001 7~0.013 3μg/μL内与吸光度线性关系良好,回归方程为y=50.549 0x-0.047 7(R2=0.999),苦参碱的平均回收率为103.17(RSD为1.70%),测得宁夏平罗县、盐池县、银川市采集的苦马豆中总生物碱质量分数分别为0.026%、0.024%、0.017%。该方法灵敏、可靠、操作简便,可用于苦马豆中总生物碱含量的测定。

摘  要:
A method was established to quantitatively determine total alkaloids in Swainsonia salsula by acid dye colorimetry.Matrine was used as a control standard and bromothymol blue as dye. The alkaloids from Swainsonia salsula and matrine reacted with bromothymol blue in the buffer solution(p H 7.6). The reaction products were extracted with chloroform. The extract was detected with ultraviolet spectrophotometer. The detection wavelength was 414 nm. The alkaloids of Swainsonia salsula collected from Ningxia province were determined by method mentioned above. The results showed that there was a good linear relationship between the absorbance of the extract and its matrine content ranged from 0.001 7 to 0.013 3 μg / μL. The regression equation established was y=50.549 0x-0.047 7(R2=0.999) with the recovery of matrine of 103.17(RSD=1.70%). The content of total alkaloids in Swainsonia salsula collected from Pinluo,Yanchi and Yinchuan was 0.026%,0.024% and 0.017% respectively. It was a highly sensitive and reproducible method to easily and quantitatively determine the content of total alkaloids in Swainsonia salsula.
