牛种布鲁氏菌SP41基因的克隆及序列分析

摘  要：
根据GenBank上发表的牛种布鲁氏菌SP41基因序列,设计合成了一对特异性引物。从牛A19疫苗中提取全基因组DNA作为模板,PCR扩增出SP41基因,连入pEASY-T3载体,测序结果表明,扩增的片段含有1 302个核苷酸,编码433个氨基酸的成熟蛋白,与已报道的布鲁氏菌2308、A-13334、S19、9-941序列的氨基酸同源性为100%;与布鲁氏菌ATCC、CCM4915、M28、M5-90、NI的氨基酸同源性为99.8%。布鲁氏菌SP41基因的克隆,为获得重组SP41蛋白及对其结构和功能的研究奠定了基础。

摘  要：
One pair of specific primers to SP41 gene was designed and synthesized based on the published sequence in GenBank.SP41 gene was cloned from genomic DNA of A19 vaccine by PCR.The product was inserted into vector pEASY-T3 to construct plasmid pEASY-T3/SP41.The sequence analysis showed that the amplified fragment was composed of 1 302 nucleotides which encoded a mature polypeptide with 433 amino acids.Its amino acid identity was 100% compared with the reported SP41 genes from the whole genome DNA sequence of 2308,A-13334,S19 and 9-941,while that was 99.8% compared with ATCC,CCM4915,M28,M5-90 and NI.The results showed that the cloning plasmid was constructed successfully,and it laid a foundation for the study of structure and function of SP41 protein.
