候鸟基因组DNA分离方法优化

摘  要:
候鸟分子生物学研究的前提是获得质量好的基因组DNA,采用标准血液DNA提取试剂盒和苯酚-氯仿法分别对采自鄱阳湖的部分候鸟血液进行基因组DNA提取。结果表明,这2种标准方法所获得DNA质量均不合要求,因此在苯酚-氯仿法基础上开展优化试验,得到优化后的苯酚-氯仿法提取条件为组织裂解液500μL、血液样品20μL、蛋白酶K 10μL和酶解时间8 h,在此条件下能分离到高质量的基因组DNA。

摘  要:
The premise of migratory bird molecular biology is to obtain high purity and good quality genomic DNA, and the DNA extraction method of common species is not suitable for the isolation and extraction of DNA from migratory birds. The first uses standard blood DNA extraction kit and classical phenol chloroform method genomic DNA blood migratory birds in Poyang Lake were extracted. The results showed that the two standard methods for the quality of DNA are undesirable. Therefore, the optimization test in the classical phenol chloroform method carried out, found that the optimized phenol chloroform method(tissue lysates 500 μL, blood samples 20 μL, proteinase K 10 μL and enzyme hydrolysis time 8 hours),the high quality genomic DNA could obtain under the condition.
