沅水五强溪水库大鳍鱯的遗传多样性分析

摘  要:
[目的]开展对沅水五强溪水库大鳍鱯(Mystus macropterus)的遗传多样性研究,了解水电开发影响下的鱼类遗传多样性.[方法]采用ISSR分子标记技术,分析洞庭湖水系沅水五强溪水库野生大鳍鱯群体的遗传多样性.[结果]8个ISSR引物在30个大鳍鱯群体样本中共获得1 162个扩增位点,其中多态位点1 141个,多态位点比例98.19%,表明五强溪水库大鳍鱯的遗传多样性丰富;水库大鳍鳠群体不同个体间的遗传距离在0.084~0.129之间,平均遗传距离为0.113,根据不同个体间的遗传距离,构建UPGMA聚类分支树状图,结果显示,30个大鳍鳠样本聚类成两个群体.[结论]沅水五强溪水库大鳍鱯群体内的遗传差异明显,至少存在两个种群的遗传分化.

摘  要:
【Objective】This study was to analyze the genetic diversity of Mystus macropterus population in Wuqiangxi Reservoir of Yuanshui River,and understand the diversity under the influence of hy-dropower development. 【Method】 Inter-simple sequence repeat(ISSR) molecular marker were em-ployed to clarify wild M.macropterus population's differentiation.【Result】The genomic DNA of the M.macropterus were amplified with 8 ISSR primers,then1 162 amplified sites were observed in 30 samples,including 1 141 polymorphic loci,and the proportion of polymorphic loci amounted to 98.19%,indicating that M.macropterus have an abundant genetic diversity. The genetic distances among individuals in population were between 0.084 and 0.129,with the average genetic distances was 0.113. The UPGMA clustering tree,based on genetic distance of 30 individuals of M.macropterus showed that they were classified into 2 populations.【Conclusion】It′s demonstrated that the intra-population′s genetic differentiation was significantly obvious in Wuqiangxi Reservoir,and M.macropterus has more than two natural geographical populations in this area.
