摘 要:
为从蛋白水平上探究ryhb对大肠杆菌酸性耐受力相关基因的影响,本研究采用非标记定量(Label-free)蛋白质组学技术对禽致病性大肠杆菌(APEC)野生株和△ryhb基因突变株进行蛋白质组学的差异性比较,并利用荧光定量PCR技术进行验证。本实验共检测到121个差异表达蛋白,其中44个蛋白的表达水平上调,19个蛋白的表达水平下调。为研究ryhb对表达蛋白的影响情况,并鉴定受其调控的相关基因,从上述差异蛋白中筛选出了hdea、hdeb、gada、gadb等与酸性耐受力相关的蛋白,并在基因水平进行验证。本研究表明,ryhb基因的缺失可能与上述4个耐酸基因转录水平上调相关,进而增强APEC耐酸性。本实验为初步探究ryhb调控通路,对APEC致病性影响提供了一定的实验依据。

摘 要:
To investigate the impact of ryhb on acid tolerance related genes in Escherichia coli at the protein level,comparison of differences in proteomics of avian pathogenic Escherichia coli(APEC) of wild-type strains and ryhb mutant strains using Label-free quantitative proteomics technology,and verified by real time PCR assay.121 differentially expressed proteins were detected,of which 44 protein expression were up-regulated and 19 protein levels were down regulated.In order to study the effect of RyhB on protein expression and identify the related genes regulated by it,the proteins related to acid tolerance,such as hdea,hdeb,gada and gadb,were screened from the above differential proteins and verified at the gene level.Finally,we drew a conclusion that the screened protein related to acid tolerance was negatively related to ryhb.This study provided a certain experimental basis for the preliminary investigation of ryhb regulatory pathway and the pathogenicity of APEC.
