基因工程小鼠肠道鞭毛虫的调查研究

摘  要:
为调查中山大学附属第一医院动物实验中心基因工程小鼠肠道感染鞭毛虫的情况,并进一步鉴定虫种,分析其对小鼠健康状况的影响。随机抽取不同来源、不同品系的基因工程小鼠的哨兵鼠采血进行血清生化分析;取盲肠组织固定于中性甲醛,进行组织切片后HE染色,比较分析鞭毛虫感染小鼠的肠道病理变化;取盲肠内容物涂片镜检,确定为阳性的样品通过ITS基因片段的PCR扩增测序,并运用MEGA 6.0软件进行种系发育分析。结果发现,抽检的小鼠全部呈鞭毛虫感染阳性;感染组的雌性和雄性小鼠血清总蛋白(TP)的含量极显著高于正常小鼠的含量(P<0.01);感染鞭毛虫的小鼠盲肠病理切片与正常小鼠盲肠相比,无明显的病理变化;获得的ITS序列与GenBank上的鼠三毛滴虫(Tritrichomonas muris)ITS序列(登录号为AY886843)同源性为84%,在种系发育树中该研究的三毛滴虫(Tritrichomonas sp.)在三毛滴虫科(Tritrichomonadidae)拓扑结构中独立形成一个自展值为86%的分支。结果表明本次分离到的三毛滴虫可能是三毛滴虫属的一个新种,并且对小鼠的基本健康无明显危害。

摘  要:
The present study investigated the infection of intestinal flagellates in genetically engineered mice and influence to the basic health of mice in the laboratory animal center of the first affiliated hospital of Sun Yat-sen University.The genetically engineered sentinel mice were randomly selected from different sources and different strains,which were used for seral biochemical analysis.In order to observe histopathological lesions of intestinal infection in mice,the cecum tissues were fixed by neutral formaldehyde for histological section.The fecal smears were observed by microscope.ITS gene sequence of positive samples were amplified by PCR and sequenced.Phylogenetic analysis was conducted by using the software MEGA version 6.0.The microscopic examination of cecal contents showed that all the mice were positive for flagellates infection.Serum total protein(TP)of female and male mice in flagellates infection group was significantly higher than that in normal mice(P <0.01).The Blast result of the ITS gene was consistent with the Tritrichomonas muris(GenBank ID AY886843)at a value of 84%.In the phylogenetic tree,the Tritrichomonas sp.of this study formed a independent branch with bootstrap value of 86% in the Tritrichomonadidae topology.It is concluded that the Tritrichomonas sp.may be a new species of genus Tritrichomonas,and it has no obvious harm to the basic health of mice.
