瞬时沉默乙醛脱氢酶对辣椒疫霉效应因子RxLR129113功能的影响

摘  要:
辣椒疫霉菌(Phytophthora capsici)产生的效应因子RxLR129113在本氏烟上的功能之一是抑制BAX引起的细胞坏死,而且已经证实乙醛脱氢酶(ALDH)是RxLR129113的互作蛋白。为了探究乙醛脱氢酶(ALDH)是否影响Rx LR129113抑制BAX引起的细胞坏死,本研究利用病毒诱导的基因沉默(VIGS)技术,克隆了乙醛脱氢酶(ALDH)基因片段,插入到烟草脆裂病毒载体(TRV)中,构建了pTRV-ALDH的VIGS载体,转入农杆菌侵染本氏烟后qrt-PCR验证成功沉默了ALDH基因。在沉默ALDH基因的本氏烟上瞬时表达RxLR129113,24 h接种BAX。结果表明,在NbALDH沉默植株上,RxLR129113仍然抑制BAX引起的细胞坏死。本研究结果表明RxLR129113与ALDH互作不影响其抑制BAX引起的细胞坏死,为进一步深入开展辣椒疫霉效应因子RxLR129113功能机制研究奠定了基础。

摘  要:
One of the functions of the Phytophthora capsici effector RxLR129113 on N. benthamiana is to inhibit BAX-induced cell necrosis, and acetaldehyde dehydrogenase(ALDH) has been shown to be an interaction protein of RxLR129113. To investigate whether aldehyde dehydrogenase(ALDH) affects RxLR129113 inhibition of BAX-induced cell necrosis, this study cloned the aldehyde dehydrogenase(ALDH) gene fragment and inserted it into tobacco using virus-induced gene silencing(VIGS) technology. In the fragile virus vector(TRV), the VIGS vector of pTRV-ALDH was constructed, and the ALDH gene was successfully silenced by qrt-PCR after agrobacterium infecting N. benthamiana.RxLR129113 was transiently expressed on N. benthamiana with silenced ALDH gene, and BAX was inoculated 24 h. The results showed that RxLR 129113 still inhibited BAX-induced cell necrosis on NbALDH-silenced plants. The results of this study indicate that the interaction between Rx LR129113 and ALDH does not affect the inhibition of BAX-induced cell necrosis, which lays a foundation for further research on the functional mechanism of P. capsici effector RxLR129113.
