纹党和红芪加工废弃物中的活性多糖对Ⅱ型糖尿病小鼠血糖血脂的影响

摘  要:
【目的】研究陇南道地中药材纹党和红芪加工废弃物中提取的活性多糖对Ⅱ型糖尿病小鼠血糖血脂的影响.【方法】将用高糖高脂喂养1个月的小鼠,以剂量为45 mg/kg腹腔注射链脲霉素(Streptozotocin,STZ)构建小鼠Ⅱ型糖尿病模型,将建模成功的小鼠随机分为正常对照组、Ⅱ型糖尿病模型组、纹党多糖(Codonopisis pilosula polysaccharides,CPP)处理组、红芪多糖(Hedysarum polybotrys polysaccharides,CHP)处理组和二甲双胍处理组.连续灌胃给药28 d,研究两种多糖对Ⅱ型糖尿病小鼠空腹血糖(fasting blood glucose,FBG)、糖化血红蛋白(glycosylated hemoglobin,HbAlc)、总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白(high density lipoprotein,HDL)、低密度脂蛋白(low density liporotein,LDL)等指标的影响.【结果】经过测定提取的纹党多糖和红芪多糖得率分别为16.52%和13.96,纯度为99.65%和99.38%,剂量在400 mg/kg时纹党多糖对正常小鼠血糖水平的降低和对Ⅱ型糖尿病小鼠治疗效果明显优于黄芪多糖.【结论】纹党多糖和红芪多糖均能降低正常小鼠的血糖,纹党多糖和红芪多糖组合降糖能力最好,能有效降低Ⅱ型糖尿病小鼠的FBG、HbAlc、TC、TG含量,促进HDL含量升高,所以纹党和红芪加工废弃物中提取的活性多糖能够显著降低Ⅱ型糖尿病小鼠血糖,并能显著改善糖尿病小鼠血脂代谢紊乱.

摘  要:
【Objective】 To investigate the effects of bioactive polysaccharides from Codonopisis pilosula and Hedysarum polybotrys produced in Longnan on glycemia and lipidemia in type Ⅱ diabetic mice.【Method】 The type Ⅱ diabetic animal model was induced with high fat diets and a single intraperitoneal injection of 45 mg/kg streptozotocin(STZ) for one month.The ICR mice were randomly divided into six groups:blank control group,positive control group,diabetic model group,C.pilosula polysaccharides(CPP) and H.polybotrys polysaccharides(CHP)-treated groups(low-,medium-,and high-dosage),metformin and adrenaline groups.The CPP and CHP were given to the mice via intragastric administration for 28 consecutive days,weeks,and then indexes of fasting blood glucose(FBG),glycosylated hemoglobin(HbAlc),total cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL) and low density lipoprotein(LDL) were measured.【Result】 The yield of extracted polysaccharides reached 16.52% and 13.96% from the waste of C.pilosula and H.polybotrys,and the purity was 99.65% and 99.38%,respectively.The 400 mg/kg·bw of CPP on the blood glucose level of normal mice and the treatment on type Ⅱ diabetic mice were better than CHP.【Conclusion】 The CPP and CHP could significantly reduce blood glucose in normal mice,could reduce the levels of FBG,HbAlc,TC and TG in serum in STZ-induced type Ⅱ diabetic mice,and increased the concentrations of HDL significantly.The CPP and CHP have effects on decreasing the blood glucose and adjusting the disorder of lipid metabolism in type Ⅱ diabetic mice.
