乳腺炎模型大鼠注射BMSCs和抗生素后血清中急性期蛋白水平变化的研究

摘  要:
【目的】探讨乳房基底部移植大鼠骨髓间充质干细胞(BMSC)对实验性乳腺炎大鼠血清中急性期蛋白水平变化的影响。【方法】通过乳头管灌注内毒素的方法建立实验性大鼠乳腺炎模型,并将70只SD雌性大鼠中的60只随机分为3组:对照组(乳房基底部注射生理盐水)、抗生素组(乳房基底部注射抗生素,5 000万单位/侧)、移植BMSCs组[乳房基底部移植BMSCs50μL(1×106个)],每组20只。试验0和1 d各屠宰5只(三组共用),之后每组于试验2、5、7和9 d分别屠宰5只,心脏采血,制备血清,检测血清中结合珠蛋白(Hp)、血清淀粉样蛋白-A(SAA)、C-反应蛋白(CRP)水平。【结果】抗生素组血清中SAA、Hp、CRP水平于2、5、7 d时均显著低于对照组(P<0.05),移植BMSC组Hp水平于2、5 d时、SAA水平于2、5、7 d时、CRP水平于5、9 d时均显著低于对照组(P<0.05)。【结论】BMSC和抗生素均能降低乳腺炎大鼠血清中急性期蛋白水平。

摘  要:
【Objective】 The purpose of this study is to investigate the impact of bone marrow mesenchymal stem cells transplanted to rats through the breast base on serum acute phase protein levels of experimental mastitis rats.【Method】Experimental mastitis model in rats was established by perfusing endotoxin through the nipple tube.60 of the 70 female SD rats were randomly divided into three groups: control group(injection of saline in breast base),antibiotic group(injection of antibiotic in breast base,50 million units / side) and BMSCs transplanted group(injection of BMSCs in breast baseμL ).There were 20 rats in each experimental group,5 of whom were slaughtered from day 2,day 5,day 7 and day 9,respectively.The rest 10 rates(control group) that were not divided into any group were killed from the first day and second day,each time 5 rats.(The data obtained from these rats would be equally compared and contrasted with data from other groups later.After that,cardiac blood was collected from their dead bodies and preserved as serum for detecting serum haptoglobin protein(HP,amylase-like protein-A(SAA) and C-reactive protein(CRP) level.【Result】Compared with data from the control group,the SAA,Hp,CRP levels in serum of the antibiotic group were significantly lower on day 2,day 5 and day 7(P<0.05).Hp level on day 2 and 5,the SAA level on day 2,5 and 7,and CRP level on day 5 and 9 of BMSCs transplanted group were much lower(P<0.05) than those of the control group.【Conclusion】 Both BMSCs and antibiotics can reduce the level of the mastitis serum acute phase proteins.
