大麦花培育种的绿苗高效再生

摘  要:
绝大多数大麦的花培绿苗分化频率极低和供试基因型之间的花培绿苗率差异极大是阻碍花培技术运用于大麦品种改良的两大难题。我们经过多年研究,已使大麦花培绿苗分化效果得到明显改善,在1991年春季接种92个F_1种F_2代的55600枚花药中,获得了2291丛绿苗。供试的F_1和F_2代花培材料中,其绿苗分化率(绿苗数/接种花药数)达50%以上的有1个,达10%以上的有7个,达5%以上的有16个,达

摘  要:
A system has been developed for efficient regeneration of green plants from harley anther cultured in vitro. Efficient regeneration of green plants was induced from anthers containing microspores at late-uninucleate stage of F_1 and F_2 materials cultured on B_5 medium containing a combination of 3.0 rag/l 2, 4-D, 1.0 mg/l NAA, 0.25 mg/l KT, 70 g/l maltose and 2.5 g/l AC. When calli were at 2mm in diameter, they were transferred to regeneration medium containing 1.0mg/l β-BA, 0.2 mg/l NAA, 25 g/l sucrose. 2291 green plants were regenerated from 55600 cultured anthers of 92 F_1 and F_2. The regeneration frequency of green plants ranged from 50% to 0.5% among different F_1 and F_2 hybrids.
