摘  要:
鸡传染性法氏囊病毒(IBDV)的VP2蛋白是该病毒的主要抗原蛋白,为制备乳酸菌黏膜免疫口服疫苗,本研究采用乳酸乳球菌(L.lactis)及其Nisin诱导的表达系统,对VP2基因进行密码子优化,构建了表达IBDVVP2蛋白的重组乳酸乳球菌L.lactis/p NZ-VP2。经正交试验对诱导条件进行优化,重组VP2蛋白(r VP2)表达量比优化前明显提高,最终得到最佳诱导条件为:OD600nm=0.5开始诱导,Nisin终浓度为2 ng/m L,诱导时间为5 h,r VP2表达量达到最大值38μg/m L。经western blot检测,r VP2能够持续性表达并具有良好的抗原性。该重组乳酸菌的构建为进一步体内免疫实验奠定了基础。

摘  要:
Avian infectious bursal disease virus(IBDV) VP2 protein is the major antigen protein,in order to prepare mucosal immunity vaccine for chicken,the codon usage biased vp2 gene was inserted into p NZ8149 vector and transformed into the Lactococcus lactis to generate L.lactis/p NZ-VP2 which expressed the recombinant VP2 protein(r VP2) by Nisin system.Under the optimized inducing conditions,i.e.the L.lactis/p NZ-VP2 was induced at OD600nm=0.5 with 2 ng/m L nisin for 5 hrs,then the r VP2 expression increased significantly up to 38.0 μg/m L.The r VP2 was detected by VP2 monoclonal antibody after induced for 3 to12 hrs.The construction of L.lactis/p NZ-VP2 lays a foundation for IBDV lactic acid bacteria immunological experiment in vivo.
