摘 要:
猴病毒 SV40 ( Simian Virus40 )在自然状态下感染恒河猴 ,导致急性病变或长期携带感染 ,影响实验用猴质量。本文研究了灭活SV40的方法 ,发现在经 1 .5mol/ L Mg Cl2 ,56℃作用 1 2 d后 ,SV40能被彻底灭活 ,灭活后的SV40不能引起细胞病变。灭活样品经测定蛋白含量为 0 .1 88mg/ m L。PCR检测表明灭活样品感染 vero细胞中 SV40 T抗原 C末端基因稳定存在 ,EL ISA检测表明灭活后的 SV40具有免疫原性。并探讨了灭活 SV40预防恒河猴感染SV40的可行性。

摘 要:
Simian virus 40 infects rhesus (macaca mulatta) natively,leading to acute pathological changes and long term latent infection,affecting the quality of rhesus used as animal models. This paper reported a simple and stable method to inactivate SV40. As a result, SV40 776 strain was inactivated by 1.5M MgCl_2 at 56℃ for 12. No typical CPE was observed on the vero cells after inoculation with the inactivated SV40 samples. The protein content in the activated SV40 sample was 0.188 mg/mL. The C end fragment of T antigen was detected using PCR in the vero cells inoculated the inactivated sample. ELISA results showed that the inactivated sample was of antigenicity. The feasibility to use inactivated SV40 in preventing SV40 infection between rhesus monkeys was also discussed.
