剑尾鱼烂鳃、烂尾病病菌的分离鉴定

摘  要:
从有明显烂尾、烂鳃症状的剑尾鱼(Xiphophorus hellerii)体表病灶处、鳃及肝脏分离出优势菌株(1201F、1201H),分别采用肌肉注射和腹腔注射方式进行人工感染实验,证实菌株1201F和1201H均能够独立引起养殖剑尾鱼发病死亡,其对剑尾鱼的半数致死量分别为2.99×107cfu/尾、1.19×106cfu/尾。经形态学观察、生理生化特性分析以及16S rDNA基因测序确定菌株1201F为柱状黄杆菌(Flavobacterium cloumnare),菌株1201H为维氏气单胞菌(Aeromonas veronii)。其中菌株1201F与柱状黄杆菌(AB078047)的亲缘关系最近,置信度为90%;菌株1201H与维氏气单胞菌的亲缘关系最近,与维氏气单胞菌(JQ795750)聚为一个分支,置信度高达93%。对24种药物的敏感实验证实菌株1201F和菌株1201H同时敏感的抗生素有阿奇霉素、诺氟沙星、恩诺沙星、左氟沙星、环丙沙星。

摘  要:
The aim of this research was to isolate the pathogens of bacterial ulceration disease in Xiphophorus hellerii and to determine their LD 50 in swordtail,and also to identify the sensitivity to antibiotics. Two bacteria strains named 1201F and 1201H were isolated from the skin,gill,and liver of dying swordtail,respectively. Both of the strains could cause the mortality of healthy swordtail independently in artificially infected experiment and were proved to be pathogens of swordtail. The LD 50 of strain 1201F to healthy swordtail by muscle injection challenge was 2. 99 × 107cfu / fish and strain 1201H was 1. 19 × 106cfu / fish by intraperitoneal injection. Based on the results of morphological observation,physiological and biochemical characteristic analysis,and 16S rDNA gene sequence,1201F was identified as Flavobacterium columnare and 1201H was Aeromonas veronii. Phylogenetic tree of Cytophaga-Flavobacterium-Bacteroides based on 16S rDNA gene indicated that strain 1201F showed the highest level of similarity to F. columnare,and the bootstrap was 90%. Phylogenetic tree of Aeromonas based on 16S rDNA gene indicated that strain 1201H was clustered closely with A. veronii( JQ795750) and the bootstrap was 93%. The antibiotic sensitivity analysis showed that both 1201F and 1201H were sensitive to azithromycin,norfloxacin,enrofloxacin,levofloxacin and ciprofloxacinum.
