红景天苷对低氧条件下培养的鸡胚肺微血管内皮细胞HIF-1α表达水平的影响

摘  要:
为观察不同浓度的红景天苷对低氧培养的鸡胚肺微血管内皮细胞(PMVECs)缺氧诱导因子-1α(HIF-1α)表达水平的影响,将原代培养的PMVECs分为5组:对照组、低氧组、低氧处理+40mg/L红景天苷(SDS)组,低氧处理+80mg/L SDS组,低氧处理+120mg/L SDS组,通过比色法检测培养细胞上清液中丙二醛(MDA)含量,应用实时荧光定量PCR(qRT-PCR)检测HIF-1α的表达水平。结果显示,与对照组相比,低氧组PMVECs上清液中MDA的含量明显上升(P<0.05),PMVECs HIF-1α的表达水平显著上调(P<0.05);与低氧组相比,PMVECs经不同浓度SDS处理后上清液中MDA的合成量显著下降(P<0.05),PMVECs HIF-1α的表达水平明显下调(P<0.05)。结果表明,SDS能有效抑制低氧处理的鸡胚PMVECs HIF-1αmRNA的表达水平,并对MDA的合成有调节作用。

摘  要:
In order to investigate the effects of salidroside on expression of hypoxia-inducible factor-1α(HIF-1α)in pulmonary microvascular endothelial cells(PMVECs)of chicken embryos cultured under hypoxic condition,the primarily cultured PMVECs were divided into five groups:control groups,hypoxia group and hypoxia +different concentrations of salidroside pretreatment groups(40,80,120 mg/L).The MDA concentration of cell culture fluid was measured by colorimetric method.The expression of HIF-1α was determined by quantitative real-time-PCR(qRT-PCR).Compared with the control group,our results showed that the concentration of MDA was noticeably increased in hypoxia group(P<0.05).Using qRT-PCR,observed that hypoxia significantly elevated the expression of HIF-1αmRNA in the cultured PMVECs(P<0.05);compared with hypoxia group,pretreatment with different concentrations of salidroside significantly attenuated hypoxia induced MDA increase(P<0.05)and dramatically decreased the production of the HIF-1α(P<0.05)in PMVECs.In conclusion,the results suggest that salidroside can effectively inhibitthe abnormal expression of HIF-1in PMVECs of chicken embryos cultured under hypoxic condition,and regulate the synthesis of MDA.
