抗虫抗除草剂转基因水稻B1C893的获得与鉴定

摘  要:
利用农杆菌介导法将Cry1Ca#和Bar基因转化水稻恢复系R893,获得6株再生植株。经过分子鉴定,发现Bar基因和Cry1Ca#基因成功整合到水稻基因组并稳定表达。测定其中单拷贝且可育的转基因株系苗期不同器官中Cry1Ca蛋白的表达量,发现根系中最低而叶片中最高,其中最高的第6号株系叶片中含量达到35.07μg/g。获得的转基因株系对除草剂草铵膦和稻纵卷叶螟均具有良好的抗性。

摘  要:
R893,a hybrid rice restorer line,was transformed with the Bar gene and the Cry1Ca#gene by the Agrobacterium-mediated method,and six plantlets were regenerated under glufosinate selection pressure. The molecular analysis of the regenerated plantlets showed that the Bar and Cry1Ca#genes were integrated into the rice genome and stably expressed. The enzyme-linked immunosorbent assay( ELISA) showed that the Cry1Ca protein expressed in different organs of the fertile transgenic plants with a single copy of exogenous gene at seedling stage. The expression of Cry1Ca protein was the weakest in root and the strongest in leaf,with the highest value of 35. 073 μg/g in leaf of the transgenic plant B1C893-6. The bioassay exhibited that the transgenic lines were of good resistance to both glufosinate and rice leaf roller.
