绵羊Klf4基因逆转录病毒载体的构建与检测

摘  要：
Klf4作为诱导产生多能性干细胞的基因之一,在体细胞的重编程过程中发挥着重要的作用。为构建绵羊Klf4基因逆转录病毒载体,参照本实验室已克隆出的绵羊Klf4基因编码区(GenBank登录号GQ280389)设计引物,将扩增的基因片段经过酶切后亚克隆到逆转录病毒载体pMXs的多克隆位点,经过酶切、PCR和测序鉴定正确后,和包装质粒pVSV-G联合转染293GP细胞获得重组逆转录病毒。通过绵羊胎儿成纤维细胞检测病毒侵染能力,并使用RT-PCR和SDS-PAGE方法检测在包装细胞和被侵染的成纤维细胞中Klf4基因的转录和表达。本试验成功获得具有侵染能力可以表达绵羊Klf4转录因子的重组逆转录病毒,为今后进一步开展以此为基础的绵羊体细胞诱导重编程研究奠定了基础。

摘  要：
Klf4 as one of the genes for inducing pluripotent stem cells,plays an important role in the somatic cell reprogramming.To construct the retrovirus vector with sheep Klf4,this experiment refered to the coding region of sheep Klf4 gene sequence,which had cloned by our laboratory(GenBank accession No:GQ280389).Then we artificially synthesized primers by the restriction enzyme digesting and connected with pMXs retrovirus vector.After identifying,recombinant plasmid was correctly detected by PCR and restriction enzyme digestion,it was cotransfected 293GP with pVSV-G plasmid.The infection ability of the recombinant retrovirus particles was detected by infecting into the sheep fetus fibroblasts,the transcription and expression of sheep Klf4 gene in infected cells was defected by RT-PCR and SDS-PAGE.Sheep pMX-Klf4 recombinant retrovirus vectors expressing Klf4 gene was construted successfully,and it could be used for inducing sheep somatic into pluripotent stem cell.
