九香虫含药血清对人结肠癌细胞SW480凋亡相关因子FADD·p53表达作用的研究

摘  要:
[目的]观察九香虫含药血清对人结肠癌细胞SW480增殖及凋亡相关因子FADD、p53表达的影响,初步探讨其诱导结肠癌细胞凋亡的作用机制。[方法]20只雄性SD大鼠按随机数字表法分为2组(生理盐水对照组和九香虫含药血清组),每组10只。其中,九香虫含药血清组给予九香虫煎剂灌胃5 d后,制备九香虫含药血清,按不同浓度体外作用于SW480细胞72 h。CCK8细胞增殖试验检测其对SW480细胞增殖的抑制作用,流式细胞术检测其对SW480细胞凋亡的诱导作用,Western blot法、荧光定量PCR法检测细胞中FADD、p53蛋白及mRNA的表达。[结果]各浓度九香虫含药血清对SW480细胞生长均有抑制作用,且明显促进其凋亡,凋亡率明显高于对照组(P<0.01)。Western bolt及荧光定量PCR结果表明,九香虫含药血清干预可使FADD表达明显增强,p53表达明显减弱。[结论]九香虫含药血清可诱导人结肠癌细胞凋亡,并影响凋亡相关因子p53、FADD的表达,从而达到抗肿瘤作用。

摘  要:
[Objective] The aim was to study the effects of the serum containing Aspongopus chinensis Dallas on proliferation of SW480(Human colon adenocarcinoma cell line) cells and expression of apoptosis-associated factors,Fas-associated death domain protein(FADD) and p53.[Methods] 20 male SD rats were randomly and equally divided into two groups of control group and Aspongopus chinensis Dallas-containing serum group,the rats in the serum group were administrated intragastrically with A.chinensis Dallas for 5 d and then the blood was collected to prepare serum containing A.chinensis Dallas,drug-containing serum was added to culture SW480 cells.Proliferation of SW480 cells was detected by cell proliferation test with CCK8.The flow cytometry was used to assay the apoptosis.The content of p53 and FADD were determined by western blot and the content of p53 mRNA and FADD mRNA by fluorescence quantitative PCR.[Results] The serum containing A.chinensis had an obvious inhibitory effect on the proliferation of SW480 for 72 hours.Drug-containing serum showed positive effect on cell apoptosis.The rate of apoptosis was higher than the control(P<0.01).The results of Western bolt and FQ PCR indicated that the Aspongopus chinensis Dallas-containing serum could intensify the FAAD expression and decrease the expression of p53.[Conclusion] The serum containing A.chinensis Dallas had a significant inhibitory effect on the proliferation of SW480.The mechanism of inducing SW480 cells apoptosis might be relevant to stimulate FADD and suppress p53.
