拂子茅组培再生体系的建立

摘  要:
为建立拂子茅植株再生体系,以茎尖为外植体,对拂子茅花叶灭菌方式以及愈伤组织诱导和植株再生培养条件进行分析与筛选。结果表明:用75%乙醇灭菌30 s,再用0.1%升汞灭菌12 min,灭菌效果最佳,去污染率达到90%;在MS+6-BA 0.2 mg/L+2,4-D 4 mg/L+蔗糖30 g/L+琼脂7 g/L的培养基上进行愈伤组织诱导,诱导率达33.4%;用MS+6-BA 2mg/L+NAA 0.2 mg/L+蔗糖30 g/L+琼脂7 g/L培养基诱导不定芽再生,诱导率最高、可达76.9%;用MS基本培养基诱导不定芽生根,生根率达到100%。

摘  要:
Plant regeneration system of Calamagrostis×acutiflora Overdam was developed using shoot tips as explants.The results showed that the suitable sterilization method was 75% alcohol in 30 s and 0.1% mercuric in 12 min.And the suitable mediums of callus induction,shoot regeneration and root culuture were MS medium with 0.2 mg/L 6-BA and 4 mg/L 2,4-D,MS medium with 2 mg/L 6-BA and 0.2 mg/L NAA,and MS basal medium,respectively.
