金毛狗脊原叶体增殖和孢子体转化条件的优化

摘  要:
为探究不同浓度的无机盐、蔗糖、KT、NAA和6-BA等对金毛狗脊原叶体增殖和孢子体转化的影响,采用正交设计实验方法,设置5因素4水平正交试验,筛选金毛狗脊原叶体增殖和孢子体转化的最佳培养基。结果表明,原叶体增殖最佳培养基为1/2MS+2%蔗糖+0.1 mg/L NAA+0.3 mg/L 6-BA+0.1 mg/L KT。孢子体转化最佳培养基为1/4MS+1%蔗糖+0.5 mg/L KT+0.1 mg/L NAA。

摘  要:
The effects of different concentrations of inorganic salts, sucrose, KT, NAA and 6-BA on the prothallus proliferation and sporophyte transformation in Cibotium barometz were investigated. The orthogonal design experimental method was used to set four levels of the above five factors to screen the best medium for the prothallus proliferation and sporophytes transformation in Cibotium barometz. Results showed that the optimal medium for prothallus proliferation was 1/2MS+2% sucrose+0.1 mg/L NAA+0.3 mg/L 6-BA+0.1 mg/L KT and the optimal medium for sporophytes transformation was 1/4MS+1%sucrose+0.5 mg/L KT+0.1 mg/L NAA.
