苹果李属坏死环斑病毒检测及其遗传多样性分析

摘  要:
为了明确李属坏死环斑病毒(prunus necrotic ringspot virus,PNRSV)在甘肃省苹果部分产区的发生及遗传变异情况,对采自5个地区表现花叶症状和无明显症状的93份苹果叶片样品进行检测,通过RT-PCR在天水和兰州苹果样品中检测到PNRSV.分别克隆了5个PNRSV分离物的外壳蛋白(coat protein,CP)基因.序列分析结果表明,5个PNRSV分离物间核苷酸和氨基酸序列一致率分别为97.3%~99.4%和95.5%~99.1%.5个PNRSV分离物与NCBI数据库中其他21个PNRSV分离物的核苷酸和氨基酸序列一致率分别为91.5%~100%和89.6%~100%.利用PNRSV CP基因序列构建了系统发育树,26个PNRSV分离物共分成3个组,本研究中的5个PNRSV分离物与美国分离物AF013287聚集在一起.基因交流分析表明,桃(Prunus persica)和巴旦木(P.dulcis)的PNRSV分离物种群之间存在频繁的基因交流,桃(P.persi-ca)和西梅(P.cerasifera)分离物之间容易发生遗传漂变.

摘  要:
To elucidate the occurrence and genetic variability of prunus necrotic ringspot virus(PNRSV)in selected apple-producing regions of Gansu Province,93 apple leaf samples with mosaic symptoms and asymptomatic samples were collected from 5 distinct areas.PNRSV was detected in Tianshui and Lanzhou apple samples by RT-PCR.The coat protein(CP)genes of five PNRSV isolates were cloned and subsequently sequenced.Sequence analysis revealed that the nucleotide and amino acid sequence identities among the five PNRSV isolates ranged from 97.3%to 99.4%and 95.5%to 99.1%,respectively.Compared with 21 additional PNRSV isolates from the NCBI database indicated nucleotide and amino acid sequence identities ranging from 91.5%to 100%and 89.6%to 100%,re-spectively.The phylogenetic tree constructed using the CP gene sequence of PNRSV categorized the 26 PNRSV isolates into three distinct groups.The five PNRSV isolates from this study clustered to-gether with the American isolate AF013287,suggesting potential phylogenetic relationships.Gene ex-change analysis indicated frequent genetic interactions among PNRSV isolates from peach(Prunus persica)and almond(P.dulcis)populations,while genetic drift was more likely to occur readily be-tween isolates from peach and prune(P.cerasifera).
