激素对蝉拟青霉胞内核苷合成的影响

摘  要:
[目的]在蝉拟青霉液体深层发酵过程中添加6-苄氨基嘌呤(6-BA)和3-吲哚乙酸2种激素以考察激素对蝉拟青霉代谢途径的影响。[方法]在蝉拟青霉液体发酵培养基中,按照5 mg/L的比例添加激素6-苄氨基嘌呤和3-吲哚乙酸,研究激素对蝉拟青霉液体发酵过程中生物量、胞内总核苷和腺苷合成的影响。[结果]2个试验组在整个发酵过程中生长代谢节奏与对照组间存在一定差异;6-BA试验组和3-吲哚乙酸试验组蝉拟青霉于发酵第3天达到最大生物量,分别为6.135 g/L和5.860 g/L,比对照组分别提高了19.10%和13.76%,显著高于对照组(P<0.05),而且试验组比对照组提前24 h达到最大值;6-BA试验组和3-吲哚乙酸试验组胞内总核苷最高含量分别达到18.49 mg/g和16.77 mg/g,显著高于对照组(P<0.05),比对照组分别提高了65.24%和49.87%;6-BA试验组胞内腺苷含量于发酵第4天达到最大值2.32 mg/g,3-吲哚乙酸试验组于第3天达到最大值1.82 mg/g,对照组于发酵第4天胞内腺苷含量达到最大值1.75 mg/g,统计分析后发现6-BA试验组胞内腺苷最高含量显著高于3-吲哚乙酸试验组和对照组(P<0.05)。[结论]2种激素在蝉拟青霉液体深层发酵过程中的添加对于提高虫草相关制品品质发挥了积极作用,同时也说明激素确实会改变微生物的代谢途径及生长节奏。

摘  要:
[Objective ]In order to study the effect of hormones on the content of nucleoside in the mycelia of Paecilomyces cicadae. [Method]6-benzylaminopurine and Indole-3-Acetic acid were added at the concentration level of 5 mg / L in the medium before the submerged fermentation.[Result]The results showed that the rhythm of mycelia growth and metabolism were different between the control group and the test groups during the fermentation process. Two kinds of hormones had an promotion effect on the biomass and the content of nucleoside of Paecilomyces cicadae.The biomass of 6-BA group and Indole-3-Acetic acid group reached to the maximum level 6. 135 g / L and 5. 860 g / L after fermentation for 3d,and were notably increased by 19. 10% and 13. 76% respectively compared with the control group( P < 0. 05),and the fermentation time could shorten to 24 h. 6-BA and Indole-3-Acetic acid could increase the content of nucleoside significantly,the content of nucleoside of test groups arrived 18. 49 mg / g and 16. 77 mg / g. In addition,the content of nucleoside were increased by 65. 24% and 49. 87% compared with the control group( P < 0. 05). The content of adenosine in mycelia of 6-BA test group and control group reached to the maximum level at 2. 32 mg / g and1. 75 mg / g respectively after fermentation for 4 days,and the content of Indole-3-Acetic acid test group arrived 1. 82 mg / g in the third day. The effect of 6-BA on the content of adenosine was better than Indole-3-Acetic acid significantly( P < 0. 05). [Conclusion] Two hormones in submerged fermentation of Paecilomyces cicadae played a positive role in improving the caterpillar fungus,at the same time also suggested that hormone could indeed change the metabolic pathway and growth rhythm of microbial.
