山兰种子无菌萌发及形态变化研究

摘  要:
研究山兰种子无菌萌发的条件,阐明种子萌发过程中的形态变化。通过比较NAA和6-BA激素组合对球茎3种发育途径影响、MS培养基盐含量和光照强度对种子萌发数的影响,得出促进山兰种子萌发的激素、培养基和光照条件;通过连续取样观察,阐明种子萌发过程中不同发育阶段的形态特征。山兰种子在黑暗条件下萌发数最高,适宜萌发的培养基为MS+NAA 0.5mg/L+6-BA 2.0mg/L,种子萌发过程分为6个阶段:胚细胞吸水膨胀阶段、心形胚阶段、鱼雷形胚阶段、子叶胚阶段、原球茎形成阶段和球茎分化阶段。

摘  要:
This paper presented the culture conditions for the asymbiotic germination of Oreorchis patens(Lindl.)Lindl. seeds and the morphology change during germination.Methods:The effcet of different ratios of 6-BA,NAA on development pathway of corm,salt strength of MS medium and light intensity on germination of seeds were observed,The morphology change was also investigated during seed germinating.Dark condition was suitable for seeds germination,the optimum medium for seeds germination was MS+NAA 0.5 mg/L+6-BA2.0mg/L,the germination process was divided into 6 stages:embryonic cell absorption water and swell stage,heart-shaped embryo stage,torpedo-shape stage,cotyledon embryo stage,protocorm formation stage and corm formation stage.
