电子束辐照翠竹幼苗的诱变效应及RAPD分析

摘  要:
对翠竹种苗的鞭根进行时长为5 s、10 s、15 s、20 s、25 s、30 s和60 s电子束辐照(剂量率10 Gy·s-1),种植后观察植株生长过程的外部形态特征并结合RAPD技术筛选突变体。结果显示:在辐照时长为5 s、15 s、30 s和60 s 4组中分别出现了叶色、叶形或株高发生显著变异的植株。统计分析显示辐照处理对植株的叶长有一定影响,其中5 s、30 s辐照组植株与对照组有显著性差异,同时各实验组的叶间距与对照相比都有显著缩短的现象。RAPD结果表明电子束辐照翠竹种苗以引起体内基因组DNA发生变异,其中5 s(条纹)组遗传变异最小,遗传距离仅为0.0280,60 s辐照处理组后代遗传变异最大,遗传距离为0.1504。此研究结果表明电子束辐照应用于翠竹诱变育种可以引起较丰富的遗传变异,为竹类产业的发展提供了一种新的育种手段。

摘  要:
The seedlings of Sasa pygmaea with rhizomes were irradiated by electron beam for5s,10s,15s,20s,25s,30s and 60s respectively( dose rate: 10Gy s- 1),after planting,their external morphological characteristics were observed during growth,and mutants were screened by RAPD analysis. The results indicated that mutant plants were significantly different in the aspects of leaf color,leaf shape or plant height for the treatments of 5s,15s,30s and 60s.Electron beam irradiation had an effect on leaf length to some extent,leaf lengths of the 5s and30s treatments were significantly different from the control,the spacing between the leaves shortened significantly in the experimental groups as well. Results of RAPD analysis indicated that the electron beam irradiation can induce variation in genome DNA of Sasa pygmaea,of which the minimum genetic distance was 0. 0280 in the 5s treatment and the maximum was0. 1504 in the 60s one. Therefore,the electron beam irradiation can be applied in mutation breeding of Sasa pygmaea,and it is a new breeding method for the development of bamboo.
