铜皂分光光度法测定猪血清中的微量游离脂肪酸

摘  要:
前言动物血液中的游离脂肪酸(FFA)含量是一项重要的生理生化指标。近来有学者认为,猪血清中的FFA含量与猪胴体瘦肉率之间有一定的相关性。为了进一步开展这方面的研究工作,需要一种灵敏、准确、快速测定血样中FFA含量的分析方法。测定血样中的FFA含量有多种方法。较早的有滴定法,目前报导较多的有气—液色谱法和铜皂分光光度法,此外还有高压液相色谱法,酶法,和放射分析法等。在这些方法中,铜皂分光光度法是一种比较简单、快速和实用的方法,适合于一般条件的实验室应用。

摘  要:
A sensitive, rapid and accurate copper soap spectrophotometric method for determination of FFA in pig serum has been reported. FFA are extracted with chloroform-heptane-methanol mixture in the presence of silicic acid so as to eliminate interference of phospholipids. Diphenylcarbazide containing 5% of diphenylcarbazone is chosen as the complex-color reagent.In the proposed method, the aqueous copper phase is denser than the organic phase containing the extracted FFA. With the organiic phase as the upper phase and the aqueous copper phase becoming the lower phase, the contamination of the aqueous copper can be eliminated.Serum sample series curves are prepared. The results shows a good agreement between the serum sample series curves and the palmitic acid standard curves. Only 0.1ml (100μl) of serum sample is required for analysis. The standard recovery of palmitic acid is from 92.5 to 105.7%. The coefficient of variation is 0.94—5.84%, which shows a good reproducibility.The proposed method is suitble for laboratories with general conditions to determine the FFA in blood sample and other biological fluids.
