Mini-Ti质粒在农杆菌菌株间的转移

摘  要:
用农杆菌介导法将外源基因导入植物是植物遗传转化的一种首选方法。为了获得较高的转化率,往往需要选择对转化目标植物敏感的农杆菌菌株。本实验利用冻融法将农杆菌LBA4404/pCG-Ⅱ中的目的片段转移到农杆菌EHA105中,通过PCR扩增、斑点杂交和测序,证明了农杆菌LBA4404/pCG-Ⅱ中的目的片段已成功转移到农杆菌EHA105中。利用该方法更换农杆菌菌株,可满足不同植物对不同农杆菌菌株的要求。

摘  要:
It is a good choice of the genetic transformation to introduce foreign genes into plants by Agrobacterium tumefaciens-mediated method. To increase the transformation efficiency, it is needed to choose the appropriate Agrobacterium tumefaciens strain for the objective plants. We have transformed the target gene of Agrobacterium tumefaciens. LBA4404/pCG-Ⅱ to the Agrobacterium tumefaciens EHA105 by freeze-thaw method, and the result was confirmed by PCR, dot blotting and sequencing. It can be satisfied the demand of different plants for different Agrobacterium tumefaciens using this method to replace Agrobacterium tumefaciens strains.
