基于高通量测序的真核微藻多样性质控分析

摘  要:
利用测序数据质量、序列聚类水平、测序数据量的合理性探讨,对辽东湾真核微藻进行高通量测序,比较不同序列聚类水平对真核微藻多样性研究的影响,以期进一步优化真核微藻的分子鉴定技术。结果发现,辽东湾17个站位每个样品获得的高质量序列数均超过87%,用于注释的序列数中碱基占比超过97%,表明测得的数据准确可靠。97%序列聚类水平对目标物种丰度的鉴定准确度较高,也会降低对物种多样性的高估。利用稀释曲线和等级聚类曲线可评估测序数据量的合理性,当曲线趋向平坦,说明测序数据量渐进合理,增加测序数据量浪费时间和成本,辽东湾的测序量无论站位评估还是组内评估均达到合理水平。根据高通量测序结果,整理了种水平辽东湾浮游植物名录、分布及特性,名录包含90种藻类,其中7种可导致鱼类死亡,25种引发过赤潮或褐潮,21种含有毒素。

摘  要:
In order to further optimize the molecular identification technology of eukaryotic microalgae, the quality control method of eukaryotic microalga diversity in Liaodong bay was discussed based on high-throughput sequencing data, such as the sequence clustering level, and the sequencing data volume. The results showed that the high-quality sequence number obtained from each sample was above 87% in Liaodong Bay, and the proportion of bases was above 97% in the sequence number which was used for annotation, indicating that the measured data was accurate and reliable. In the 97% sequence clustering level, the identification of species abundance was more accuracy, and also reduced the overestimation of species diversity. The reasonableness of sequencing data volume was evaluated by using dilution curve and hierarchical clustering curve. When the curve became flat, it indicated that the sequencing data volume was reasonable, so it was not nessary to increase the amount of sequencing data. Furthermore, the sequencing quantity of algae in Liaodong Bay reached a reasonable level no matter on station evaluation or group evaluation. According to the results of high-throughput sequencing, the study compiled the list, distribution and characteristics of phytoplankton in the Liaodong Bay. The list contained 90 kinds of algae, including 7 fish-kill algae, 25 red tidealgae or brown tidealgae, and 21 toxinalgae.
