低温环境下南海深海沉积物中可培养细菌的多样性及其抑菌活性分析

摘  要:
模拟低温(10℃)环境分离纯化培养南海深海沉积物中的细菌,基于16S rRNA基因序列对南海深海沉积物可培养细菌多样性进行分析,并利用滤纸片扩散法测试可培养细菌发酵液的抑菌生物活性。结果表明,从南海深海沉积物中共分离获得相关可培养细菌32株,分属于3个菌门(变形菌门、厚壁菌门和拟杆菌门)的19个种,其中以芽孢杆菌为优势菌群。分离获得的南海深海沉积物可培养菌株中有17株的代谢物对大肠杆菌、金黄色葡萄球菌、枯草芽孢杆菌和蜡状芽孢杆菌4种指示菌均表现出抑制作用,占分离菌株种类的89.5%。说明低温环境下对深海沉积物可培养细菌多样性和活性均存在一定影响。

摘  要:
The marine bacteria were isolated from the deep-sea sediment of the South China Sea,and cultivated at low temperature of 10℃ using conventional culture-dependent method. Then the diversity of cultivable bacteria was investigated and analyzed based on 16 S rRNA gene sequence. Meanwhile,the bacteriostatic activity of fermentation broth of cultivable marine bacteria was determined by filter paper-agar diffusion method. The results showed that,a total of 32 cultivable marine strains were isolated from the deep-sea sediment of the South China Sea,belonging to 19 species of 3 phyla( Proteobacteria,Firmicutes,Bacteroidetes),of which,Bacillus strains were dominant. Furthermore,the metabolites of 17 strains had bacteriostatic activity against Escherichia coli,Staphylococcus aureus,Bacillus subtilis and Bacillus cereus,which accounted for 89. 5% of all isolated bacterial species. It can be deduced that low temperature has a certain influence on diversity and activity of cultivable marine bacteria isolated from the deep-sea sediment of the South China Sea.
