棉蚜保幼激素环氧水解酶基因克隆及表达初步分析

摘  要:
保幼激素环氧水解酶(Juvenile hormone epoxide hydrolase,JHEH)在保幼激素(Juvenile hormone,JH)的代谢途径中起重要的降解作用。为初步分析JHEH在棉蚜(Aphisgossypii Glover)生长发育中可能存在的功能,本研究通过转录组测序并结合RACE技术,克隆AgosJHEH基因,其开放阅读框为1401 bp,编码466个氨基酸,预测蛋白质分子量为52.6kDa,等电点8.19。N末端存在由20个氨基酸组成的疏水性信号肽序列,存在催化三联体Asp(230)、Glu(408)、His(437)氨基酸残基以及阴氧离子洞Tyr(304)、Tyr(378)和HGWP花样结构氨基酸残基,与豌豆蚜氨基酸序列有很高的同源性。RT-qPCR结果表明AgosJHEH在棉蚜无翅若虫3、4龄表达量显著低于其他龄期,在无翅成虫中各生命时期没有显著差异,有翅成虫腹部显著低于翅中的表达量。本研究首次对AgosJHEH进行结构分析及功能预测,为深入理解其对棉蚜JH滴度的调控机制及开发新型杀虫剂奠定基础。

摘  要:
Juvenile hormone epoxide hydrolase(JHEH) is considered one of the most important enzymesin degraded juvenile hormone(JH).In this paper,Agos JHEH was cloned from Aphis gossypii Glover by using the RNA-seq transcriptome dataset and RACE strategy in order to investigate the function of JHEH in A.gossypii growth and development.The open reading frame was 1401 bp long,and encoded 466 amino acids.Its predicted molecular mass and isoelectric point was 52.6 kDa and 8.19,respectively.According to signal peptide prediction,Agos JHEH had a hydrophobic signal sequence with 20 amino acids in N-terminal region.The residues Asp(230),Glu(408),His(437) that form the potential catalytic triad,and the residues Tyr(304),Tyr(373),and the HGWP motif that constitute the potential oxyanion hole were found in the deduced amino sequences.The Agos JHEH amino acid sequences have high homology to JHEH from Acyrthosiphon pisum(Harris).We also used real-time quantitative PCR to test the expression level of Agos JHEH in different tissues,and life stages of A.gossypii.The results showed that Agos JHEH gene was expressed throughout nymph stage to adult stages,the expression level was significantly lower in nymph of 3 rd,4th in star than those of other in stars,while expression levels in adult were not significantly different.The expression level in abdomen of alatae adult was significantly lower than the expression level of wing.This is the first report on analyzing structure and predicting function of Agos JHEH.Our findings lay the foundations for understanding the regulatory mechanism of A.gossypii JH titer and are helpful for discovery of new insecticides.
