摘 要:
采用直接测序法对7种鲤(Cyprinus carpio):德国镜鲤(C.carpio L.mirror)、松荷鲤(C.carpio L)、松浦鲤(C.carpio var.Songpu)、易捕鲤(C.carpio var.Yibu)、大头鲤(C.pellegrini T.)、松浦镜鲤(C.carpio var.Songpu mirror)和野鲤(C.carpio haematoperus)TLR9基因的SNP位点的多态性和遗传多样性进行了研究。结果显示,在292个个体中共检测出37个碱基变异位点,均位于基因外显子区域,占整个片段长度的0.84%,其中20个SNP位点为非同义突变。野鲤的碱基突变位点最多,松浦镜鲤的碱基突变位点最少(P<0.05),其他各品种间无显著性差异(P>0.05);野鲤的非同义突变位点最多,松浦镜鲤的非同义突变位点最少(P<0.05),其他各品种间无显著性差异(P>0.05)。稀有等位基因频率大于0.05时,经Hardy-Weinberg卡方检验结果显示各品种中除了德国镜鲤和松浦镜鲤外,大多数位点都处于平衡状态(P>0.05);各品种的PIC值处于中低度多态水平(PIC<0.5)。卡方检验分析结果显示,各品种间共有24个位点的基因型频率存在显著性差异(P<0.05),其中11个位点为非同义突变位点,这可能是影响TLR9基因免疫应答功能的原因。

摘 要:
According to TLR9 gene sequence of the Cyprinus carpio: C. carpio L. mirror,C. carpio L,C. carpio var. Songpu,C. carpio Yibu,C. pellegrini T.,C. carpio var. Songpu mirror,C. carpio haematoperus,the SNPs of the TLR9 polymorphism gene and genetic diversity were analyzed by direct sequencing method in seven different carp groups. Results of the study was showed that 37 base mutations loci had been detected in exon region in 292 individuals,accounting for 0. 84% of the whole length of the fragment,and of which 20 SNP loci were non-synonymous mutations. The polymorphic loci and non-synonymous mutation loci in all the seven species were detected. The results showed that C. carpio haematoperus had the most polymorphic loci and non-synonymous mutation loci,and Songpu mirro carp had the least ones( P <0. 05),there were no significant different among the other species( P > 0. 05). The Hardy-Weinberg detecting results showed that the majority polymorphic loci were balanced except C. carpio L. and Songpu mirro carp( P > 0. 05). The PIC values were in the low level of polymorphism( PIC < 0. 05). Chi-square analysis showed that the genotype frequency of 24loci had significant different in each group,including 11 non-synonymous mutation loci( P < 0. 05). This may be the reason influenting the main function of TLR9,leading to its immune response.
