一株鸽副粘病毒分离株的毒力及免疫原性

摘  要:
从病/死鸽中分离到1株病毒,血清学鉴定该分离病毒为禽Ⅰ型副粘病毒(PPMV);该病毒经毒力测定结果显示,PPMV的毒力与鸡新城疫病毒(NDV)La Sota株的毒力相似,为自然弱毒株;对该分离株F基因裂解位点的序列分析结果表明,PPMV F裂解位点附近的氨基酸序列为112G-R-Q-G-R-L117,属于弱毒株的特征性序列。将该株病毒制备成灭活疫苗,分别免疫鸽、鸡,结果都能诱导鸽、鸡体产生较高水平的HI抗体;分别以鸡新城疫标准强毒株F48E9、本组分离于鸡的野毒株CPMV(MDT 45.4 h,ICPI 1.85,IVPI 2.42)和分离于鹅的野毒株GPMV(MDT 59 h,ICPI 1.88,IVPI 2.39)攻击免疫鸡群,免疫鸡群可得到保护,而非免疫鸡群被攻击后全部致死。结果表明,用该分离毒制成的油乳剂灭活苗对鸡安全,具有良好的免疫效果,可抵抗鸡新城疫、鹅副粘病毒的攻击。用HI方法检测该株病毒与La Sota株之间的交叉血凝抑制试验,结果显示La Sota株与PPMV两毒株间的抗原性差异较小。

摘  要:
One strain of paramyxovirus typeⅠ(from pigeon,named as PPMV) was isolated and identificated serologically.The results of virulence test indicated that the PPMV showed similar pathogenicity to La Sota strain of NDV,which was avirulent to chickens naturally.The results of amino acid sequence analysis showed that the sequence around the cleavage site of the fusion protein of PPMV was 112 G-R-Q-G-R-L117,which is typical motif of avirulent type.After inactivated,the isolate was inoculated to chickens and pigeons,respectively.High level of HI antibody against the isolate was detected.Then the immunized chickens were challenged by F48E9 strain of NDV and the two field strains of CPMV,GPMV(PMV was solated from chickens and geese respectively and preserved by ourselves.).The results showed that all of the immunized chickens could be protected.Therefore,we were sure that the inactivated oil vaccination against PPMV was safe and effective to chickens against challenge of GPMV and NDV.Crossing test of HI showed that there was little difference on antigenicity between PPMV and La Sota strain of NDV.
