摘 要:
[目的]为凤桥镇通过合理利用氟烷基因提高猪肉品质奠定理论基础。[方法]从嘉兴县凤桥镇的规模化养猪场随机抽取种猪血样,通过PCR-RFLP技术进行氟烷基因检测。[结果]HhaⅠ内切酶的识别位点为5′GCG↓C3′。PCR扩增产物是含RYR1基因第1 843位点长度为659 bp的片段。如果猪的基因型为NN,由于未发生胞嘧啶C到胸腺嘧啶T的突变,可以看到2条带。如果猪的基因型为nn,由于发生了胞嘧啶C到胸腺嘧啶T的突变,只能看到1条带。如果猪的基因型为Nn,可以看到3条带。在长白猪、杜洛克猪和大约克猪3个供试品种中没有检测出氟烷基因隐性纯合子(nn),其n等位基因频率分别为2.5%、1.9%和1.6%。[结论]建议在凤桥镇利用的氟烷基因杂合子(Nn)的公系和氟烷基因显性纯合子(NN)的母系开发猪的胴体优势。

摘 要:
[Objective] The purpose was to lay a theoretical foundation for enhancing pork quality through utilizing halothane gene reasonably in Fengqiao town.[Method] The blood samples were extracted from breeding pigs randomly in the large-scale piggeries of Fengqiao town in Jiaxing County and the halothane gene was detected by PCR-RFLP technique.[Result] The recognition site of endonuclease Hha Ⅰ was 5′GCG↓C3′.The PCR amplified product was a segment containing the 1843rd site of gene RYR1 and its length was 659 bp.If the genotype of pig was NN,there would be 2 bands appeared for that there was no mutagenesis from cytosine C to thymine T occurred.If the genotype of pig was nn,there would be only 1 band appeared for that there was mutagenesis from cytosine C to thymine T occurred.If the genotype of pig was Nn,there would be 3 bands appeared.There was no allozygote(nn) of halothane gene detected out in the 3 tested varieties of landrace pig,duroc pig and yorkshire pig and their frequences of allele gene n were 2.5%,1.9% and 1.6% resp.[Conclusion] It was suggested that the carcass advantage of pig should be developed by using male lines with heterozygote(Nn) of halothane gene and female lines with dominant homozygote(NN) of halothane gene in Fengqiao town.
