新疆细毛羊leptin基因第二、三外显予扩增片段的多态性分析

摘  要：
根据leptin基因在GenBank中的已知序列设计两对引物,采用PCR-SSCP和DNA测序技术在新疆细毛羊群体中进行单核苷酸多态性(SNPs)检测分析.结果表明,新疆细毛羊leptin基因第二、三外显子都具有多态性.其中,第二外显子扩增片段上有AA和AB两种基因型,AA基因型为优势基因型;而在第三外显子有CC、CD两种基因型,以CC基因型为优势基因型.并且经过DNA测序发现,在leptin第一内含子上有两处突变.即1TrG三个连续碱基的插入和C→T的碱基替换;而在第三外显子上发生了M替换突变.

摘  要：
According to the known sequence of leptin gene from GenBank, two primers were designed. The study used gene sequencing and PCR-SSCP assay to analyze the single nucleotide polymorphisms (SNPs) of the Xinjiang fine-wool sheep. The results showed that both exon 2 and exon 3 of leptin in Xinjiang fine-wool sheep were of the polymorphism. Two genotypes AA and AB were detected on the amplified fragment of the exon 2, of which the genotype AA was dominant. And two genotypes CC and CD Were detected on the amplified that three leptin. fragment of the exon 3, the genotype CC was dominant in it. Using gene sequencing assay, we found consecutive bases TI'G inserted and C / T mutated in intron 1 and G / T mutated in exon 3 ofleptin.
