应用正交设计建立三岛柴胡愈伤组织快速繁殖体系

摘  要:
[目的]建立三岛柴胡愈伤组织快速繁殖体系。[方法]采用正交优化设计的方法,以细胞平均增长量为参考指标,对三岛柴胡愈伤组织的基本培养基、初始pH值、6-BA、NAA及2,4-D的添加量进行筛选优化。[结果]5种因素对试验结果的影响均达到显著水平。基本培养基中,N6对试验影响最大;初始pH值为5.6有利于细胞的生长;3种激素中,2 mg/L 6-BA和0.5 mg/L NAA以组合的方式加入培养基中时,可获得了较高的细胞鲜重。通过对试验结果的分析比较,得出三岛柴胡愈伤组织细胞生长最有利的培养基配方为:N6+6-BA 2 mg/L+NAA 0.5 mg/L,pH为5.6;2,4-D对三岛柴胡愈伤组织的生长有一定的抑制作用。[结论]该研究结果为三岛柴胡愈伤组织的进一步扩大培养和悬浮体系的建立提供了试验数据和理论支持。

摘  要:
[Objective] To establish B.falcatum callus rapid propagation system.[Method] With the reference to the average growth of cells,the media,initial pH value,6-BA,NAA and 2,4-D of B.falcatum callus were optimized by orthogonal optimal design method.[Result] The five factors had significant influence on the test results,especially N6,the initial pH of 5.6 was conducive to the growth of cells;of the three hormones,the largest cell fresh weight can be obtained when the combination of 6-BA(2 mg/L) and NAA(0.5 mg/L) was added to the medium.The most advantageous medium for B.falcatum callus cell growth was N6+2 mg/L 6-BA + 0.5 mg/L NAA,pH 5.6.The cell growth of B.falcatum callus was inhibited by 2,4-D when it was added in medium.[Conclusion] The study results provide experimental data and theoretical support for further expanding the culture and establishing the suspension system of B.falcatum callus.
