猪痢疾短螺旋体荧光PCR检测方法的建立与初步应用

摘  要:
在对设计合成的引物和6-carboxy-fluorescein(6-FAM)荧光素标记的Taq Man水解探针进行筛选的基础上,通过反应体系优化,建立了猪痢疾短螺旋体荧光PCR检测方法。特异性、敏感性、重复性及临床样品的检测结果均表明建立的方法快速、特异、灵敏,整个检测过程(包括样品的前处理时间)可在4 h内完成。建立的方法大大缩短了该病原的检测周期,并为临床诊断提供快速、敏感、特异性强的检测方法。

摘  要:
Based on the screening of the designed primers and TaqMan hydrolysis type probes labeled with 6-carboxy-fluorescein(6-FAM) reporter dyes,this research developed a real-time PCR detection method for Brachyspira hyodysenteriae by means of optimazation of reaction conditions.The sensitivity,specificity,repeatability and detection clinical samples of the real-time PCR were examined respectively.The results showed that the method were rapid,specific and sensitive.The assays could be finished in 4 hours(including the pretreatment of samples).The developed method shortens the detecting period of swine dysentery,and at the same time it provides a rapid,sensitive and specific method for clinical diagnosis.
