摘 要:
为寻找家蝇防治新靶点提供新的思路,研究灭多威作用于家蝇胚胎细胞系MDEC-07114细胞后对细胞色素C和Caspase 3的影响。用4 mg/mL灭多威干预MDEC-07114后,分别于加药后2、6、12、24 h收集细胞,用蛋白质印迹法(Western blot)检测细胞色素C和Caspase 3的表达。结果表明,细胞色素C随作用时间的延长释放增多,Caspase 3随作用时间的延长活性增强。根据试验结果,推测灭多威可能是通过损伤线粒体膜,引起细胞色素C的释放,进而激活Caspase 3,发生蛋白酶级联反应,从而导致MDEC-07114细胞凋亡。

摘 要:
In order to provide new ideas for preventing and controlling Musca domestica, effects of methomyl on Cytochrome C and Caspase 3 in cell line(MDEC-07114) of Musca Domestica embryo were studied. With the intervention of 4 mg / mL methomyl, MDEC-07114 cells were collected after exposing to methomyl for 2 hours, 6 hours, 12 hours and 24 hours, re- spectively. The release of Cytochrome C and Caspase 3 in MDEC-07114 was detected with western blot. The results showed that the release of Cytochrome C increased and the activities of Caspase 3 were stronger with the increase of time. Based on the experiment results, it is deduced that methomyl may cause the release of Cytochrome C by mitochondrial damage, then activate caspase cascade and induce the apoptosis in MDEC-07114 cells.
