O型口蹄疫病毒不同宿主适应毒P1基因分析

摘  要:
为了明确O型口蹄疫病毒不同宿主适应毒P1基因变异趋势,将口蹄疫病毒分别接种于牛、乳鼠、猪及BHK-21细胞,获得相应宿主的适应毒,以各宿主适应毒RNA为模板,扩增出P1(1A、1B、1C、1D)基因,将各适应毒株扩增的P1基因序列相互比对。结果表明,牛、乳鼠、猪、BHK-21细胞宿主适应毒株P1基因的核苷酸及其编码的氨基酸序列均未发生缺失;部分关键性氨基酸发生了变异,变异发生在VP1主要抗原区域的141-160位和200-213位、VP2抗原表位的156-166位和217-220位;VP3抗原蛋白发生较少氨基酸变异;VP4均未发生变异。试验结果为进一步分析不同宿主适应毒基因组的变异关系奠定了基础。

摘  要:
In order to identify P1 gene sequence differences of different host-adaptive type O foot-and-mouth disease virus(FMDV)and study the trend of P1 genetic variation,FMDV was inoculated into cattle,neonatal mice,swine and BHK-21 cell line to obtain corresponding adapted virus.The RNA of FMDV in each adapted virus was taken as template for reverse transcription and amplification of P1(1A,1B,1C,1D)gene,the sequences of each adapted virus amplified P1 gene were compared with each other.The results were as follows:different host-adapted virus nucleotide and amino acid sequences were no deletions;the mutation occurred in some of the key amino acids,the variation occurs in 141-160 and 213-200 in the main antigen region of VP1,156-166 and 217-220 in the antigen epitope region of VP2;less variation occurs in antigen protein region of VP3;no mutation occurs in region of VP4.The results provided important information for genome analysis of different host-adapted FMDV.
