高效液相色谱仪检测鸡组织器官喹烯酮残留方法的建立

摘  要:
建立用高效液相色谱法测定鸡组织中喹烯酮浓度的方法,用于监测鸡组织器官中喹烯酮的残留。方法:采用Prodigy 5μPhenyl-3色谱分析柱,Waters TM486可变波长紫外检测器,甲醇-水(70:30)为流动相。组织样品用乙酸乙酯、乙腈、正已烷、氯仿等试剂提取处理纯化后进样,在312 nm波长处检测,流速为1.0 mL/min。标准曲线的线性范围:脂肪、肾脏和肌肉均为0.005-0.080μ/g;肝脏为0.005-0.500μ/g。方法平均回收率:肌肉为(99.23±2.75)%;脂肪为(105.15±8.65)%;肝脏为(93.55±11.16)%;肾脏为(104.10±9.80)%。精密度考察喹烯酮的日内各组织各浓度的RSD(相对标准差)均小于5.1%,日间测得各组织各浓度的RSD均小于8.5%。本方法灵敏度高,重复性好。

摘  要:
A HPLC method is described for the analysis of quinocetone in chicken tissues. Prodigy 5 μ Phenyl-3 column (4. 6 mm±250 mm 5 μ micron) was used. The mobile phase consisted of methanol-water (70 : 30). A flow rate of mobile phase is 1. 0 mL/min. A waters model 480 instrument was used throughout the experiment. The absorption wavelength of determination is 312 nm. Cure regression equation: fat,Y=4±10-5X-0. 0293, R2=0. 9944(n=5). The ranges of quinocetone calibration curves were 0. 005-0. 080 μg/g;liver,Y=3±10-5X - 0. 02,R2 = 0. 9926(n=8). The ranges of quinocetone calibration curves were 0. 005-0. 500 μg/g;kidney,Y= 4±10-4X-0. 0394,R2 = 0. 9933(n=10). The ranges of quinocetone calibration curves were 0. 005-0. 800μg/ g;muscle,Y=6±10-5X-0. 0631,R2 = 0. 9926(n=6). The ranges of quinocetone calibration curves were 0. 005 -0. 080 μg/g. The lowest detectable concentration of quinocetone was 0. 005 μg/g for fat , liver, kidney and muscle. While the average recovery of the method was (99. 23 ±2. 7)% in muscle; (105. 15 ±8. 65)% in fat; (93. 55 ± 11.16)% in liver;l04. 10±9. 80% kidney. The RSD of intra-day of all concentrations and all tissues was all less than 5. 1%,those of inter-day of all concentrations and all tissues was all less than 8.5%, The method is higher sensitivity and good repeat.
